Vasonite is a xanthine derivative. Improves microcirculation in areas of impaired blood supply. Improves the rheological properties of blood (fluidity) by affecting the pathologically altered deformability of erythrocytes, increases the elasticity of erythrocyte membranes, inhibits the aggregation of erythrocytes and platelets and reduces the increased viscosity of blood.
The mechanism of action of the drug is associated with the inhibition of phosphodiesterase and the accumulation of cAMP in the cells of vascular smooth muscles and in the blood cells. Pentoxifylline inhibits the aggregation of platelets and erythrocytes, reduces the level of fibrinogen in the blood plasma and enhances fibrinolysis, which leads to a decrease in blood viscosity and an improvement in its rheological properties.
Improves the supply of oxygen to tissues in areas of impaired blood circulation (including in the limbs, central nervous system, and to a lesser extent in the kidneys). With occlusive lesion of the peripheral arteries (intermittent claudication) leads to an increase in walking distance, elimination of night cramps, calf muscles, and a decrease in pain at rest.
In case of impaired cerebral circulation, it improves symptoms. It has a weak myotropic vasodilating effect, slightly reduces the systemic vascular resistance and slightly dilates the coronary vessels.
Circulatory disorders in: atherosclerosis, diabetes, Raynaud's disease;
treatment of trophic ulcers, treatment of cerebral circulation disorders.
Application during pregnancy and lactation
The use of the drug Vazonit during pregnancy is contraindicated.
If necessary, use of the drug during lactation should stop breastfeeding.

From the digestive system: possibly - nausea, vomiting, diarrhea, dry mouth, decreased appetite, feeling of fullness in the abdomen, pain in the epigastrium, increased activity of liver enzymes (ALT, AST, LDH) and alkaline phosphatase, cholecystitis, hepatitis, jaundice; rarely - unpleasant taste in the mouth, hypersalivation.
From the side of the central nervous system: relatively rarely - headache, dizziness, anxiety, sleep disturbances, convulsions.
From the senses: rarely - conjunctivitis, scotoma, blurred vision, ear pain.
From the side of the cardiovascular system: when using the drug in high doses, it is possible to decrease blood pressure, tachycardia, angina pectoris, cardiac arrhythmias, flushing of the facial skin, "hot flashes" of blood to the skin of the face and upper chest, edema.
Allergic reactions: possibly - itching, skin rash, urticaria, Quincke's edema; rarely - anaphylactic shock.
From the hematopoietic system and hemostasis: rarely - bleeding (from the vessels of the stomach, intestines, skin and mucous membranes), thrombocytopenia, leukopenia, pancytopenia, hypofibrinogenemia, changes in the peripheral blood picture, aplastic anemia. In this regard, it is necessary to carry out regular monitoring of the blood picture.
Others: rarely - malaise, increased brittle nails, changes in body weight, nasal congestion.
Pentoxifylline can enhance the effect of drugs that affect the blood coagulation system (indirect and direct anticoagulants, thrombolytics), antibiotics (including cephalosporins - cefamandol, cefaperazone, cefotetan), valproic acid.
Increases the effectiveness of antihypertensive drugs, insulin and oral hypoglycemic drugs. Cimetidine increases the plasma concentration of pentoxifylline (risk of side effects).
Co-administration with other xanthines can lead to excessive nervous excitement in patients.
How to take, course of administration and dosage
Enteric-coated tablets are swallowed whole with a little water.
The daily dose is divided into 3 doses. The initial dose is 600 mg / day.
As the condition improves, the dose can be reduced to 300 mg / day.
Prolonged dosage forms are prescribed 2-3 times a day.
Symptoms: nausea, dizziness, decreased blood pressure, fever (chills), tachycardia, drowsiness or agitation, loss of consciousness, areflexia, possible tonic-clonic seizures, vomiting of the "coffee grounds" type (as a sign of gastrointestinal bleeding), arrhythmias.
Treatment: gastric lavage, then symptomatic therapy is carried out. There is no specific antidote. In case of vomiting with traces of blood, gastric lavage is unacceptable.
Treatment should be carried out under the control of blood pressure. In patients with diabetes mellitus taking hypoglycemic drugs, administration in large doses can cause severe hypoglycemia (dose adjustment is required).
When administered simultaneously with anticoagulants, it is necessary to carefully monitor the indicators of the blood coagulation system. In patients who have undergone recent surgery, systematic monitoring of Hb and hematocrit is necessary.
The administered dose should be reduced in patients with low and unstable blood pressure. In the elderly, dose reduction (increased bioavailability and decreased elimination rate) may be required.
The safety and efficacy of pentoxifylline in children is not well understood. Tobacco smoke can reduce the therapeutic efficacy of the drug. The compatibility of the pentoxifylline solution with the infusion solution should be checked on a case-by-case basis.
